Description
| Application | The GenElute™ HP Plasmid Miniprep Kit offers a fast and efficient solution for high-throughput plasmid preparation from E. coli cultures. This kit combines silica-based membrane technology with the convenience of vacuum manifold or automated liquid handling workstation format. Following lysis the DNA is bound to the silica membrane and contaminants are removed with two simple wash steps. The DNA is then eluted in water or TE buffer by centrifugation. The recovered plasmid DNA is ready for immediate use in applications such as restriction enzyme digestion, sequencing, PCR, and transfections. |

| Principle | An overnight recombinant E. coli culture is harvested with centrifugation and subjected to a modified alkaline-SDS lysis procedure followed by adsorption of the plasmid DNA onto silica in the presence of high salts. Vacuum wash steps then remove contaminants. Finally, the bound plasmid DNA is eluted in water or Tris-EDTA buffer. |
